Neo-Epitope Prioritization

Neo-epitopes are important targets for individualized cancer immunotherapy. Recent advances in next generation sequencing and bioinformatic approaches to predict immunogenicity of neo-epitopes have improved target selection for therapy. However, in many cases only a fraction of predicted epitopes generate a specific T-cell response. Therefore, various assay techniques such as ELISpot, MHC-binding, or mass-spectrometry are being applied to confirm and prioritize neo-epitopes for individualized therapies. The ability to create meaningful and truly patient-individualized sets of neo-epitopes ultimately requires high-throughput formats and thus large numbers of small scale, high quality, and low cost peptides, delivered rapidly.

Immunopeptidomics applies mass spectrometry to discover, verify, and prioritize neo-epitopes. In contrast to other assays, it can be largely paralleled and analyzes many peptides at the same time. However, challenges such as limited sensitivity and specificity still exist. Peptides quickly synthesized at low costs have proven to be excellent reference standards for HLA peptides. NeoSpikeMixes are customized small scale peptide libraries to be used as references for HLA peptides by mass spectrometry. They provide fast and cheap access to reference peptides to increase accuracy of neo-epitope identification.
