What Are Peptide Pools?

A peptide pool is a mixture of multiple synthetic peptides combined into a single experimental reagent. Instead of testing every peptide separately, researchers can use peptide pools to investigate many peptide sequences simultaneously within one experimental condition.

The composition of a peptide pool depends on the scientific question. Pools may contain overlapping peptides spanning an antigen, selected immunodominant epitopes, peptides arranged into matrix subpools, peptides selected to represent sequence diversity, or reference peptides for mass spectrometry-based applications.

For cellular immunology, peptide pools are widely used for antigen-specific T-cell stimulation and screening. Other pool formats support antigen discovery, immunopeptidomics or targeted proteomics. The appropriate design therefore depends on the assay, target and required downstream analysis.

T-Cell Research

PepMix™ Peptide Pools

PepMix™ pools are designed for antigen-specific T-cell stimulation in functional assays such as ELISpot, cytokine staining, cell-mediated cytotoxicity and proliferation assays. They are also used for immune monitoring, T-cell expansion, cellular immune-response profiling and T-cell epitope identification.

More than 500 ready-to-use PepMix™ peptide pools are available, including pools for viral, bacterial and fungal antigens as well as tumor-associated antigens.

Antigen Discovery

SpotMix Antigen Discovery Pools

SpotMix pools contain small-scale peptides generated using JPT's SPOT technology. The peptides are produced in an automated process and purified by dialysis before pooling.

They are designed for broad antigen target discovery, T-cell epitope discovery and biological screening. SpotMix PLUS can additionally support neo-epitope-based immune-monitoring projects.

Overlapping Peptide Pools

Overlapping peptides can span an antigen sequence, enabling broad representation of potential epitopes for T-cell stimulation, screening and immune-response analysis.

Matrix Pools

Peptides are distributed across multiple subpools so individual reactive peptides can be identified from the pattern of positive pools. Matrix pools are also used within JPT Epitope Mapping Peptide Sets.

Selected Epitope Pools

Selected peptide pools combine published immunodominant epitopes. JPT examples include CEF and CEFX control pools and Pan Select PepMix™ pools.

ULTRA Pools

ULTRA pools are designed to represent sequence diversity within viruses or cancer-related targets using a reduced number of peptides while maintaining broad sequence coverage.

A peptide pool is a mixture containing multiple synthetic peptide sequences in one experimental reagent. Pooling allows researchers to expose a sample to many peptides simultaneously instead of testing every peptide in a separate condition.
Depending on their composition, peptide pools can be used for antigen-specific T-cell stimulation, immune monitoring, epitope and antigen discovery, biological screening, immunopeptidomics and targeted proteomics.
An overlapping peptide pool contains peptides that collectively span a protein or antigen sequence with neighboring peptides sharing part of their sequence. This format is commonly used when broad representation of potential epitopes across an antigen is required.

Matrix pools distribute peptides across several subpools so that each peptide occurs in a defined combination of pools. Comparing positive subpools can help identify the peptide responsible for an observed response. JPT also uses matrix pools within Epitope Mapping Peptide Sets.
For antigen-specific T-cell stimulation and immune monitoring, start with PepMix™. For broad antigen or T-cell epitope discovery, consider SpotMix. NeoSpikeMix supports immunopeptidomics applications, while SpikeMix™ is designed for targeted proteomics. For specialized layouts such as matrix or custom antigen-spanning pools, JPT can configure a custom peptide pool.
Each vial contains 25ug (or 15 nmol) of each of the individual peptides. However, in some cases we also offer larger amoun(200µg, or 120 nmol, of each peptide). Please refer to the product datasheet provided on the product page.

We guarantee presence of all peptides, high accuracy of aliquotation and batch-to-batch reproducibility.
For SpikeTides TQ and SpikeTides TQL, the tag carries a moiety enabling the quantification of the target peptide during HPLC-runs using specific wavelengths. The tag is optimized to enable efficient cleavage using standard proteases like trypsin. After cleavage, the proteotypic peptide with the native C-terminus is available for the MRM assay.

The cleavage of the SpikeTides can be performed in the same assay solution as the tryptic digest of the target protein takes place.

JPT Peptide Technologies has validated the cleavage efficiency in many conditions. The efficiency of the cleavage can be checked by doing HPLC-runs after the cleavage was performed. The signal of the cleaved tag should be visible at a retention time of 0 to 0.5min (C18, 3µ,, 20x2mm, 1ml/min flowrate, 6 minute gradient, 5% to 95% Acetonitril water) and should light up when analyzed at 350nm.
