About PepMix™ Human CMV (pp65)

The PepMix™ Human CMV (pp65) peptide pool offers a highly effective tool for studying immune responses to Human Cytomegalovirus (HCMV). The peptides included in this pool represent T-cell epitopes that are recognized by CMV-specific CD4 and CD8 T-cells across a broad range of MHC contexts (see table below). These T-cells, when activated, typically produce pro-inflammatory cytokines such as interferon-gamma (IFN-γ), interleukin-2 (IL-2), and tumor necrosis factor (TNF).

Pool of 138 peptides derived from a peptide scan (15mers with 11 aa overlap) through 65 kDa phosphoprotein (pp65) (Swiss-Prot ID: P06725) of Human Cytomegalovirus (HCMV) - strain AD169.

Applications of PepMix™ Human CMV (pp65)

Detection and Functional Analysis

The PepMix™ Human CMV (pp65) antigen is a powerful tool for:

  • Detecting T-cell responsiveness to HCMV, a common issue during HCMV infection.
  • Using as a positive control in T-cell stimulation assays due to its ability to stimulate CMV-specific T-cells effectively.

Isolation and Enrichment of T-Cells

This product facilitates the isolation of CD4 and CD8 T-cells from PBMCs (Peripheral Blood Mononuclear Cells) through activation-dependent processes. By leveraging antibody-labeled magnetic beads, HCMV pp65-specific T-cells can be enriched and expanded, enabling the creation of T-cell lines or clones for in-depth studies.

Advanced Applications

JPT’s PepMix™ Human CMV (pp65) is ideal for high-quality cell assays such as:

  • Cytokine production-dependent assays, including ELISPOT and flow cytometry.
  • Detection of surface activation markers such as anti-CD154 or anti-CD137.

Unlike non-specific controls like staphylococcus enterotoxin B (SEB) or phytohemagglutinin (PHA), the PepMix™ pool ensures T-cell receptor-dependent activation, providing a more physiological and reliable solution for peptide-based T-cell activation assays.

About PepMix™ Peptide Pools

PepMix™ peptide pools are protein-spanning mixtures of overlapping peptides designed for a variety of immunological applications, including:

  • Antigen-specific T-cell stimulation in assays like ELISPOT, ICS, cell-mediated cytotoxicity, or proliferation assays.
  • In-vitro T-cell expansion.
  • Dendritic cell pulsing.
  • Immune monitoring and profiling of cellular immune responses.
  • T-cell epitope identification.
  • Standardization of T-cell assays.

Benefits of PepMix™ Peptide Pools

  • Guaranteed identity and purity for each peptide.
  • Certificate of Analysis (CoA) and HPLC-MS data provided.
  • High batch-to-batch consistency.
  • No false positive T-cell responses due to contaminating deletion peptides.
  • No toxic inhibition of T-cell responses thanks to stringent peptide purification.
  • Minimal endotoxin contamination through low bioburden processes.
  • Animal-derived component-free (ADCF) policy.
  • HLA-independent stimulation using antigen-spanning peptide pools.

Additional Insights into HCMV and pp65

Role of CMV pp65 in Viral Biology

The pp65 protein, a critical tegument protein of HCMV, plays an essential role in:

  • Viral replication by aiding the incorporation of viral proteins into the host cell.
  • Modulating the host cell’s immune responses to ensure effective immune evasion.
  • Regulating HCMV-encoded genes during viral genome expression.

Research Applications

Understanding the role of cellular proteins and HCMV genes in the context of pp65 provides insights into:

  • The mechanisms of immune evasion by HCMV.
  • Development of antiviral strategies targeting pp65.
  • Broader implications for studying opportunistic infections in immunocompromised individuals.

Testimonials
"To establish a novel role for myeloid derived suppressor cells (Pallett et al, Nat. Med. 2015) in chronic viral infection, we utilised the PepMix CEF Pool (extended) as well as a custom synthesized PepMix spanning the core region of HBV genotype D . Whereas, the CEF peptide pool consists of 32 peptides, each corresponding to a defined HLA class I-restricted T-cell epitope from Cytomegalo, Epstein-Barr, and Influenza virus, the latter custom PepMix included 15meric peptides overlapping by 10 amino acids. Specifically this composition enabled us to monitor both the antiviral CD8+ and CD4+ T cell responses in chronic HBV infection and the non-antigen specific T cells that are known to mediate immunopathology in the liver. Our entire experience with JPT, from ordering/delivery to use in the lab was excellent. Not only were the reagents able to perform reliably and consistently in vitro from batch to batch, the customer and technical support provided was continually available and efficient when needed. JPT will remain our go-to company for purchasing peptides."
