What is a Peptide Affinity Tag?
One of the major challenges the production of recombinants is the purification of the target protein from other components expressed by the organism. To circumvent this, a recombinant protein is usually expressed with an additional amino acid sequence that allows for a specified subsequent affinity purification. Most common are the
- Histidine tag: A string of histidine residues that binds to metal ions, often used for affinity chromatography.
- FLAG-tag: A short sequence that binds to specific antibodies, ideal for detection and purification.
- Strep-tag: Binds to streptavidin for high-affinity purification.
Once a tag is bound to the affinity resin, the protein is released, typically on an HPLC column. The release is facilitated by the excess addition of a competitive agent of the affinity tag.

Here are some widely used peptide affinity tags, each serving unique purposes in protein purification, detection, and study:
- FLAG Tag: A small octapeptide (DYKDDDDK), this tag binds to anti-FLAG antibodies, enabling purification and detection through immunoaffinity methods.
- His Tag: A polyhistidine tag (usually 6-10 histidine residues) allows for purification via nickel or cobalt affinity chromatography.
- Strep Tag (II): a small octapeptide (WSHPQFEK) that binds specifically to streptavidin or its derivatives, allowing for high-affinity purification.
