Radioligand therapy (RLT) – as the name implies – makes use of a radioactive cationic metal (i.e. 177Lu, 68Ga, and 64Cu), bound to a chelating agent such as DOTA (tetraacetic acid) or NOTA (1,4,7-Triazacyclononane-1,4,7-triacetic acid) attached to a peptide, which typically target a somatostatin or other endocrine receptor. For a decade, macrocyclic peptides became valuable ligands thanks to their targeting and PK properties.
