Pool of 28 peptides derived from a peptide scan (truncational scan; peptides from 11 to 17aa lengths) through Spike glycoprotein - Fusion peptide AA814-830 (Swiss-Prot ID: P0DTC2: 814 - 830) of SARS-CoV-2 (Severe Acute Respiratory Syndrome-related coronavirus 2).

Amount: 25 ug (approx. 15 nmol) / peptide (for stimulation of up to 2,5 x 108 cells)
Purity: crude (major peak by LC-MS is guaranteed to be peptide of interest for each peptide)

Background: A 15 amino acid stretch of the SARS-CoV-2 spike glycoprotein was recently shown to induce strong T-cell reactivity in individuals following SARS-CoV-2 vaccination and infection, and responses were also found in individuals with prior exposure to other human coronaviruses (Loyal et al., Science 2021). The amino acid stretch (named iCope) was independently predicted as a region of major antigenic potential in silico (Ong et al. Comp. Struct. Biotechnol. J. 2021).

The respective peptide sequence is located in the spike glycopeptide fusion domain and is likely to give rise to one or more universal immunodominant coronavirus-specific peptide epitopes across a range of presenting HLA-alleles.

Our PepMix peptide pool spans the region from SARS-CoV-2 Spike aa814 to aa830 including all possible 17-, 16-, 15-, 14, 13-, 12- and 11-amino acid peptides in order to maximize T cell stimulation.

"To establish a novel role for myeloid derived suppressor cells (Pallett et al, Nat. Med. 2015) in chronic viral infection, we utilised the PepMix CEF Pool (extended) as well as a custom synthesized PepMix spanning the core region of HBV genotype D . Whereas, the CEF peptide pool consists of 32 peptides, each corresponding to a defined HLA class I-restricted T-cell epitope from Cytomegalo, Epstein-Barr, and Influenza virus, the latter custom PepMix included 15meric peptides overlapping by 10 amino acids. Specifically this composition enabled us to monitor both the antiviral CD8+ and CD4+ T cell responses in chronic HBV infection and the non-antigen specific T cells that are known to mediate immunopathology in the liver. Our entire experience with JPT, from ordering/delivery to use in the lab was excellent. Not only were the reagents able to perform reliably and consistently in vitro from batch to batch, the customer and technical support provided was continually available and efficient when needed.
