Pool of 138 peptides derived from a peptide scan (15mers with 11 aa overlap) through 65 kDa phosphoprotein (pp65) (Swiss-Prot ID: P06725) of Human Cytomegalovirus (HCMV) - strain AD169.

PepMix™ Human CMV (pp65) peptide pools are essential tools for studying the immune response to CMV pp65, a major Cytomegalovirus protein critical in viral pathogenesis and immune recognition. This unique peptide pool stimulates T-cell responses, targeting both CD4 and CD8 T-cells across diverse MHC contexts (refer to table). These activated T-cells typically release pro-inflammatory cytokines, including interferon-gamma (IFN-γ), interleukin-2 (IL-2), and tumor necrosis factor (TNF), which play critical roles in modulating immune defenses against hCMV infection.

The pp65 protein, a prominent tegument protein in Human Cytomegalovirus (HCMV), plays an integral role in viral replication, immune evasion, and host cell infection. PepMix™ Peptide Pool Human CMV pp65 Applications

The PepMix™ Human CMV (pp65) antigen detects T-cell responsiveness to HCMV. As most people have CMV-specific T-cells recognizing HCMV pp65 the PepMix™ Human CMV (pp65) is often used as a positive control in T-cell stimulation assays. It is also useful for isolating CD4 and CD8 T-cells from PBMCs after activation. This isolation uses antibody-labeled magnetic beads. HCMV pp65-specific T-cells can be enriched through peptide stimulation and proliferation, creating T-cell lines or clones. JPT’s PepMix™ Peptide Pool Human CMV (pp65) is suitable for various cell assays needing high-quality peptides, including cytokine production-dependent assays like ELISPOT or flow cytometry, and detection of surface activation markers such as anti-CD154 or anti-CD137. Unlike non-specific controls such as as staphylococcus enterotoxin B (SEB), phorbol-12-myristate 13-acetate(PMA)/Ionomycin, or phytohemagglutinin (PHA), PepMix™ provides a T-cell receptor-dependent activation control, making it a more physiological and relevant option for peptide-based T-cell activation assays.

HLA Specificty of PepMix™ Peptide Pool Human CMV pp65

Peptides from the CMV pp65 protein are known to be presented by a wide range of HLA-molecules (note that some studies reported low-resolution, serologic specificities whereas others reported specific alleles). JPT can provide individual T-cell stimulating CMV-derived pp65 peptides for each mentioned allele. MHC molecules presenting peptides derived from human CMV-pp65 (see table).

Med. 2015) in chronic viral infection, we utilised the PepMix CEF Pool (extended) as well as a custom synthesized PepMix spanning the core region of HBV genotype D .
