Quality Control in Custom Peptide Synthesis

Quality control is a cornerstone of our custom peptide synthesis process. After synthesis of peptides, each peptide undergoes routine analysis by LC-MS, MALDI-MS, or HPLC to confirm its identity and peptide purity. JPT provides a thorough analytical data sheet and corresponding raw data documentation with each order. All documentation is meticulously double-checked and archived before leaving our facility. Our standard quality control includes LC-MS with ion trap, quadrupole, or TOF detection, MALDI-MS, or HPLC. We also offer a variety of optional Peptide Analysis Services, including Stability Testing, Peptide Content Determination, and Cell Toxicity Testing:

  • Amino acid analysis (AAA)
  • Peptide sequence analysis via mass spectrometry
  • Residual solvent and water determination
  • Solubility and stability testing
  • Toxicity, sterility, endotoxin, and bioburden testing
  • NMR (nuclear magnetic resonance)
  • And many more!
For more QC/QA options tailored to clinical applications, please see our Clinical Peptides and Pools.

Modifications and Labels for Custom Peptide Synthesis

JPT also excels in the design and production of custom peptides with various modifications, such as fluorescent labels, biotinylation, alkylation, glycosylation, phosphorylation, and more. These modifications improve peptide functionality and stability, broadening their applications in research and medicine.

Our customization options include:

  • Unnatural & unusual amino acids
  • Isotope-labeled peptides
  • Macrocyclic peptides and post-translational modifications
  • Fluorescent & FRET peptides
  • Biotinylated & tagged peptides
  • Long, cyclic, or dimeric peptides
  • Various linkers, spacers, & PEGs
  • Protein conjugates & immunogenic peptides

The Science Behind Custom Peptide Synthesis

Peptide synthesis involves the creation of peptides by sequentially adding amino acids to a growing chain. This process can be performed using either solid-phase peptide synthesis (SPPS) or liquid-phase peptide synthesis (LPPS). SPPS, developed by Robert Bruce Merrifield, is the most commonly used method due to its efficiency and versatility. JPT adds an additional step, called capping, after each amino acid coupling. Capping prevents side reactions and deletion sequences, which can lead to false positive results. For more information, please watch our capping video:

In addition to classic solid phase custom peptide synthesis, JPT uses SPOT synthesis, which allows the synthesis of hundreds of micro scale peptides in parallel. The resulting high-throughput peptides are fast and have the lowest custom peptide synthesis prices on the market.

Testimonials

Testimonials for Custom Peptide Synthesis

"The RV 144 HIV trial is considered as one of first successful HIV vaccine trials. It has become clear that the V2 loop of gp120 is an important site for immunogenicity and protection from HIV infection. The use of JPT's PepStar™ microarray technology has been very useful for the correlation of the clincial outcome with humoral immune responses. As have the cyclic peptides been from JPT to validate these findings!"
J. Currier, PhD, Walter Reed Army Institute of Research, Rockville, Maryland, USA

"Our research relies heavily on developing robust high-throughput screens with fluorescent peptides. We have found that JPT’s are the best on the market because the signal-to-noise ratio is very high, providing the sensitivity we need for the screens. Their peptides always perform well. In addition, the knowledge, wonderful customer support, and fast turnaround time provided by JPT have been invaluable in helping us develop the best peptides for our assays."
Prof. Carla Koehler, UCLA, Los Angeles, CA
